リンパ球増殖性疾患の悪性化におけるIL-6と癌遺伝子変異

研究概要

1.ヒトbcl-2に対するモノクローナル抗体の作成
IgMクラス133種、IgGクラス136種作成した。IgGクラスの方がaffinity強く、Western blotにてbcl-2α及びγ両分子結合ばかりであることを確認した。
2.抗bcl-2抗体を用いた単離細胞の染色
単離細胞をホルマリンで固定し、サポニンにて透過性を高めた後抗bcl-2抗体を用いてindirect immunostainingを行ない、FACSにて解析した。B細胞株を用いて検索したところ従来市販のものよりも強いaffinityを示す抗体、II-526とII-177を見出した。
3.染色特異性の検討
本抗体による細胞染色の妥当性を胸腺細胞を用いて検討した。胸腺細胞は大別して、CD4^-8^-,CD4^+8^+,CD4^+8^-,CD4^-8^+に分けられ、CD4^+8^+においてbcl-2の発現低下が生じるといわれている。本抗体において、bcl-2陽性細胞はCD4^-8^-では98%、CD4^+8^+では79%、CD4^+8^-では99%、CD4^-8^+では100%で、mean fluorescence intensityもCD4^+8^+のみ低下を示した。これらの結果より、本抗体はbcl-2発現検索に有用であることが明らかである。
4.扁桃Bリンパ球のbcl-2発現
扁桃リンパ球を汎BCD19と活性化Bにて発現するCD38を用いて分画し、その各々の細胞内bcl-2発現を検討した。その結果CD38^-では陽性であったが、CD38^+では陰性であった。即ち、活性化B細胞になるとbcl-2のdown-regulationが生じると考えられる。
5.IL-6の骨髄種細胞株内bcl-2発現に対する影響
RPMI-8226はIL-6に無反応の骨髄種株であるが、bcl-2を強く発現している。そこでIL-6を加えて培養しbcl-2発現量の変化を検討した。その結果、全くbcl-2の発現には変化が認められなかった。
