ヒトがん抑制遺伝子のp53の細胞生物学的機能の解析

研究概要	我々の本年度の研究目的はヒトがん抑制遺伝子p53の細胞生物学的機能を解析することであった。そのために、Mutant p53(mp53)をヒト正常細胞、および、不死化細胞(腫瘍性はない)にトランスフェクトし、そのmp53の作用を検討し、以下の結果を得た。 1)細胞の形態的変化を誘導しなかった。 2)mp53を正常ヒト細胞に移入しても染色体の変化はなかった。 3)細胞の増殖亢進や細胞密度の上昇はみられなかった。 4)正常細胞にmp53を移入しても細胞の不死化はみられなかった。 5)このmp53を移入した細胞を、10^<-6>Mの4NQOで処理すると不死化した。同条件の4NQOの処理のみでは細胞は不死化しなかった。この事実はmp53が正常ヒト細胞を不死化させ易くすることを示している。即ち、p53の変異に化学発癌剤による変異が細胞におこれば、ヒト細胞において癌化の初期段階に必須である細胞の無限増殖性がおこることをみいだした。 6)不死化した細胞にmp53を移入しても腫瘍化しなかった。 7)以上の事実は、mp53は細胞の腫瘍化より、むしろ、不死化に働くことを示している。人体の種々の異なる組識に発生する多くの腫瘍にp53の変異が認められている。それぞれの分化度の異なる細胞の癌化にp53がジェネラルに働いているとすれば、その共通する事柄、すなわち細胞の不死化にp53が作用している可能性がある結論される。
