c-myc遺伝子プロモーターに結合する転写因子ZF-5の細胞増殖抑制効果

研究概要

1.c-myc遺伝子の転写におけるZF5の機能。
c-myc遺伝子プロモーターをCAT(chloramphenicol acetyle transferase)遺伝子の上流に結合したレポーター(c-myc CAT)を用いたCAT assayでは、ZF5はCATの発現を抑制した。すなわち、ZF5はc-myc遺伝子の転写抑制因子である。
2.その他の遺伝子に対するZF5の作用。
Herpes simplex virus thymidine kinase(HSV-tK)遺伝子のプロモーターに対するZF5の作用を、CAT assayで解析したところ、やはり抑制的に働いた。一方、human immunodeficiency virus(HIV)LTRに対しては転写活性化機能を示した。このように、ZF5はプロモーター特異的に全く相反する作用をなすことが明らかになった。
3.ZF5自身の構造及び機能解析。
HSV-tkのプロモーターを用いて、ZF5の機能に関わる領域の同定を試みたところ、ZF5のN末端に近接した62個のアミノ酸領域に強い転写抑制効果が認められ、ZF5のDNA結合領域(zinc finger)には、その機能は認められなかった。一方、HIV-LTRのプロモーターに対してはzinc fingerは転写活性化作用を示した。このように、ZF5の各領域の機能にもプロモーター特異的な相違が認められた。
4.ZF5の生物学的作用
テラトカルシノーマP19細胞のretinoidによる分化誘導に伴って、alternative splicingに基づくと思われるZF5 mRNAの質的変化が認められた。
