研究概要	本年度はこれまでに我々の研究室で分離し貯蔵しているハムスター由来のBHK21細胞株の細胞増殖に関する温度感受性変異株、tsBN2,tsBN7、tsBN67、tsBN250、tsBN462を用いて研究を行い、主な成果としては次の事が挙げられる。1)アポプトーシス抑制変異遺伝子の分離。昨年度の成果に基づきアポプトーシスを起こす他の変異が我々の持っている温度感受性変異株の中に、あるか調べた。その結果、相当数の変異がアポプトーシスを高温で起こすことが判明した。その中で、最も顕著なDNA分子の切断像を示すtsBN269を取り上げその変異を相補するヒト遺伝子の分離を開始した。既に、ヒトDNAを持つ第一次形質転換株が分離されており、現在、第二次形質転換を行なっている。2)新たなG1期進行必須遺伝子のクローニング。高温でG1期停止をするtsBN67変異株を相補するヒト遺伝子の分離を進めているが、逐に変異を相補するヒト遺伝子DNA断片を分離するのに成功した。現在、2kbのORFを持つcDNA断片が分離されており、これが変異を相補するか否か調べている。cDNAの塩基配列が正しいとするとこれまでに分離されていない新しいタンパク、それも特殊な構造をしたタンパクをコードしているようである。分離されたcDNAはX染色体の遺伝子に由来していた。3)tsBN7遺伝子DAD1の解析。DADIタンパクの機能の消失がアポプトーシスを起こしたことを証明するため、DAD1を多量に発現したときに他の細胞系、ならびに線虫の系のアポプトーシスが現実に抑制されるか否かを調べている。4)CCG1遺伝子の機能。この遺伝子産物がTBP結合タンパクのTAF250であることが他に研究者の報告により判明した。我々はtsBN462株がCCG1に変異を持つことを、変異部位を決定することにより確認した。さらに、この株が高温でアポプトーシスを起こすことがわかった。
