血液幹細胞に特異的に発現されるTIE受容体のリガンドの検索

研究概要	(1)血液幹細胞のin vitro増幅が可能になると、骨髄移植や遺伝子治療を大きく進展させると思われる。我々は、新規の受容体型チロシンキナーゼ(RTK)TIEを解析し、それらの結合因子を同定できれば、将来、幹細胞の試験管内増幅も可能になると考え、以下の検討をおこなった。FACSにより純化した血液幹細胞から、TIEおよびTEK RTK遺伝子をクローニングした。両者は約50%の相同性をしめし、細胞外は、免疫グロブリン、EGF、フィブロネクチン様ドメインよりなり、細胞内には、短いinsertionをもつキナーゼドメインがみられた。両者ともに、血液幹細胞に選択的に発現され、分化とともに発現が低下するのが認められた。RT-PCR法による幹細胞からのチロシンキナーゼ遺伝子のクローニングは、有効でこのほかにも、METファミリーに属すると考えられるSTK/RONやCSKにホモロジーを示すHYLなどの新しい遺伝子が同定された。 (2)TIE遺伝子にFLAGをつなぎ、細胞外ドメインからなる可溶性TIE蛋白を発現させた。これを用いて、抗FLAG抗体により、リガンドを発現しているとおもわれるストロマ細胞との結合を検討しているが、いまだ有意の結合を確認していない。今後、TIE-免疫グロブリンキメラ遺伝子の作成、中和活性をもつTIE抗体の作成が必要と考えられる。これと平行して、ES細胞のin vitro gene targetingなどにより、幹細胞由来の新規のチロシンキナーゼ遺伝子の造血における役割を、明らかにしたい。全体として、構造解析は、進んだが、機能解析は、緒についたばかりである。
