研究概要

c-mybプロトオンコジーン産物(c-Myb)は特異DNA配列に結合する転写活性化因子である。c-Mybの中央部に存在するロイシンジッパー構造を壊すと、c-Mybの転写活性化能、細胞がん化能が顕著に上昇することから、この部位にインヒビターが結合することを推定していた(Kanei-Ishiiら、PNAS,1992)。本年度はc-MybをプローブにしてcDNA発現ライブラリーをスクリーニングし、インヒビターのcDNAクローニングを得た。Lassin(leucine zipper associated inhibitor)と名付けたインヒビターは68kDの蛋白質で3つのロイシンジッパー構造を持ち、LassinがMybに結合するとMybのDNA結合が阻害されることが明らかとなった。またLassinはMyb以外の転写因子にも作用する可能性が示唆された。一方Mybはロイシンジッパーを介してダイマーを形成し、ダイマーはDNAに結合ができないことが示され、Myb自身がインヒビターとして作用するnegative autoregulationの機構があることも明らかとなった。
