研究概要	1 効率的な不斉空間を構築するキラルリチウムアミドを設計して、環状アキラルケトンの不斉アルキル化反応を実現、触媒化に成功した。キラルリチウムアミドの溶液構造を^6Liおよび^<15>NNMRにより観測し、実際の反応との相関性に関する知見を得た。また、新たなタイプの塩基として設計、合成したキラルなリチウム2-アミノアルコキシドが、フェニル酢酸エステルのマイケル付加反応の不斉誘起に有効であることがわかった。 2 フェニルアラニン誘導体の不斉α-アルキル化反応は、α-位炭素の不斉を唯一の不斉源として69〜88%eeで進行する。本不斉誘導はmixed aggregateメカニズムによるものではなく、中間体アニオン種自体が持つキラリティーによるものであることがわかった。 3 糖質を不斉源として有機合成反応に利用することを考え、グリコシドの二つのアノマーを簡便に作り分けることを目的として研究を行った結果、遊離の糖と各種の求核剤から触媒的に効率良くグリコシドを得る手法を開発することができた。 4 反応性に富む有機金属反応剤を炭素求核剤として組み込み可能な不斉触媒反応系の構築を目指して検討を進めた。その結果、立体化学と反応性の制御を同時に可能とする有機銅の不斉付加をキラルホスフィンアミドを配位子として実現した。また、複数の反応点を有する不飽和イミンの位置選択性の発現要因を分子軌道法計算から明らかにした。さらに、従来合成困難であったキラルアミノエーテル配位子の合成法を開発できた。 5 アリル亜鉛化合物のオレフィン付へのエナンチオ選択的付加に初めて成功すると同時に、その反応遷移状態を分子軌道法計算によって明らかにした。またフラーレンを置換基として持つ光学活性リン化合物を合成し、これによって不斉合成が行えることを初めて明らかにした。 6 受容体の立体構造が既知の場合に、薬物結合部位に安定に結合できる構造を既存化合物の三次元データベースから検索する方法Adαm&Eveの開発に成功した。市販試薬のデータベースであるACD-3D (MDL社:約11万化合物) を対象に、HIV proteaseの系に適用してみた。ヒットした構造のうち12個の化合物を入手し活性を測定したところ、5個に既知のノンペプチド阻害剤ハロペリドールよりも強い活性が認められた。
