研究概要	A1)ヘム蛋白質についての、時間分割ラウエ法及びNMR分光法による研究(森島) Leu29置換ヒトミオグロビンに関しては、新たにLeu29Ser置換体を調製した。Wild typeに比べて、k(on)が1/15に減少した。親水性になったことにより、水分子が進入したためと考えられる。 A2)リン酸化合物を結合する酵素についての、時間分割ラウエ法、酵素化学による研究(小田、西野、倉光) グルタチオン合成酵素:caged-ATP:遷移状態アナログ(TSA)の三元複合体について、ラウエ法測定を行った。フラッシュ前と直後(45秒)について測定した。その結果、ATPのγリン酸がTSAに移ることを確認出来た。(小田)複合金属フラビン酵素であるキサンチン酸化酵素につき、結晶化は温度に依存し、21℃が最適であることがわかった。(西野)アミノ基転移酵素の基質となるフェニルピルビン酸と2-ケトグルタル酸のcaged化合物を合成した。(A03公募、古田との共同)酸性条件で十分遅い反応を見るために、高度好熱菌Thermus thermophilus由来のアスパラギン酸アミノ基転移酵素の大量発現系を確立した。(倉光) A3)加水分解酵素についての時間分割ラウエ法及び酵素化学による研究(三井、上杉、田之倉) RNaseA結晶へのcaged化合物の溶解度が低い問題に悩まされていたが、今回、コハク酸アンモニュウムを使用することにより、良好な結晶を得た。硫酸アンモニュウム結晶とコハク酸アンモニュウム結晶とでは、格子定数に数%の差があるだけであるが、後者のほうが、はるかに白色X線に耐性であることを発見した。分子間相互作用の様相の違いとの関連を追求中である。(三井)2',3'-サイクリックヌクレオチドに対応するcaged化合物の合成は極めて難航している。(上杉)ペプシノゲンの結晶をハンギングドロップ法で作製しラウエ法による測定を行った。アルミニュウムのフィルタを入れることによって、かなり良好なスポットが観測されるようになった。(田之倉) A4)蛋白質構造のダイナミックスを追求するための新しいNMR法の開発(楯) 緩和時間分データから、psec領域での蛋白質の主鎖の局所運動性に相違があることを示した。カルボニル炭素の緩和パラメーターから、水素結合状態など、数msec程度での時間域での構造安定性に直接かかわる量を抽出する手法を開発した。 A5)計算機実験に基づく蛋白質の動的構造解析(北尾) ピコ秒より遅い時間オーダーで起こっている運動が、時間分割ラウエ法で観測されるようなミリ秒かそれより遅い運動とどのように関係しているのか、今後更に解明していく。
