Gunnラットへの遺伝子導入による先天性黄疸の解明と治療の基礎研究

研究概要	GunnラットはUDP-glucuronosyl transferaseの欠損によりビリルビンの抱合不全をおこし、先天性の黄疸を呈するヒトのCriegler-Najjar症候群のモデル動物である。私達は、ビリルビンを抱合するUDP-glucuronosyl transferaseラットcDNAをクローニングするとともに、生体組織への効率のよい遺伝子導入法HVJ-liposomeを開発したので、これらを用いて成育Gunnラットへの肝臓へ遺伝子導入を行ない、黄疸の遺伝子レベルでの治療の可能性について検討した。 HVJ-liposome法によりUDP-glucuronosyl transferaseのcDNA約40μgをGunnラットの肝臓に直接導入し、3日後に酵素活性を測定すると正常ラットの約10%の酵素が発現された。遺伝子導入後、3日毎にラット血中のビリルビン値を測定すると、導入3〜4日後から減少し、正常に比べ約30%の減少を示し、これが1回の投与で約30日持続した。30日後に2度目の投与を行なうと、さらに約1ケ月約30%減少を認めた。一方、Gunnラットは新生仔期に小脳へのビリルビン沈着により小脳障害が併発する。そこで生後4日のGunnラットの肝臓に、HVJ-liposomeによりUDP-glucuronosyl transferase cDNAを導入すると生後25日目で約30%の血中ビリルビン値の減少を認めると共に、64%の小脳重量の回復を認めた。しかし、正常ラットの小脳重量と比較するとまだ十分な回復とはいえない。UDP-glucuronosyl transferaseは1つの遺伝子からスプライシングの相違により種々のtranscriptがつくられることが知られており、私達は現在用いているcDNAより、強力なビリルビン抱合能をもつと思われる新たなcDNAをクローニングした。このcDNAの遺伝子導入により、Gunnラットの黄疸の治療は、さらに改善されると期待される。
