組換えを起こしやすい反復配列に結合するタンパクの分離

研究概要	真核生物のゲノム中には多くの種類の縦列型反復配列が存在する。それらは染色体のセントロメア領域やテロメア領域に大量に存在するが、染色体に散在している反復配列もある。ミニサテライトやマイクロサテライト等がそのよく知られた例である。縦列型反復配列は相同な配列が隣接しているわけだから、組換え反応を起こし易いという特色を持つ。反復配列に見られる組換え反応の生物学的意義や分子機構を知るために、我々はGGAという3塩基が繰り返す反復配列を研究対象としてきた。それは2本の2重鎖DNA鎖がグアニン:グアニン結合し、複合体形成をとり易いという特色がある。 (CCT/GGA)nのようにHomopurine:homopyrimidine鎖が繰り返す配列は、triplex構造やcruciform構造などの特異な構造をとることが報告されている。これらの構造は、組換え反応の中間体の一つと考えられるので、その構造特性を検討した。(GGA/TCC)n 配列は、環状および線状DNAでS1ヌクレアーゼに感受性をもつ。(GGA/TCC)n配列を含むプラスミドDNAを合成ヌクレオチド(GGA)11と、インキュベーションすると、電気泳動で遅れる見かけ上3本鎖の会合分子ができあがる。その構造を、DMS(ジメチル硫酸)処理、DEPC修飾(一本鎖プリン塩基を特異的に修飾)、オスミウム酸修飾(一本鎖ピリミヂン塩基を特異的に修飾)、DNase I消化を用い、検討した。3薬物処理では、CCT鎖が顕著に修飾された。これはG:Gペアリングにより2本のGGA鎖が結合し、CCT鎖が一本鎖構造をとることを示唆している。また、この複合体はtriple strandに見られるDNase I抵抗性を示さなかった。従って、(GGA/TCC)n反復配列はtriplex構造とは異なった構造をとると考えられる。一方、Guanine-richなGGA鎖はテロメアと類似構造を持つので、テロメアに見られる逆平行のG:G pairingを示すかどうかを検討した。その結果、テロメアとは異なる平行の結合をすることが分かった。この特異な構造体の意義については今後の課題として残っている。
