研究概要	1.神経系細胞の動的な細胞基質間接着構造とアクチン及びアクチン結合蛋白質神経細胞とアストロサイトにおけるアクチンフィラメント・ゲルゾリン・アクトミオシン系蛋白質(ミオシン・カルデスモン・トロポミオシン)の細胞内局在を検索した。その結果、神経成長円錐にゲルゾリンとカルデスモン・トロポミオシンがアクチンフィラメントとともに局在していることを見い出した。この所見は、成長円錐のCa依存性の活動性にゲルゾリンおよびアクトミオシン系収縮要素が関与していることを示している。また、培養アストロサイトがアクチンフィラメントを豊富に含む活発に運動する基質接着構造を形成することを明らかにした。この接着構造には、ゲルゾリンとアクトミオシン系蛋白質が集積しておりその運動性の物質的基盤となっていることが示唆された。 2.神経細胞におけるアネキシンVI結合蛋白質神経細胞膜骨格のCa制御機構を明かにすることを目的として細胞膜直下に局在することが知られているCa結合蛋白質アネキシンVIの標的蛋白質の検索をラット脳の細胞骨格分画を用いて行った。その結果、アネキシンVIは、細胞膜骨格主要構成員カルスペクチンとCa及びリン脂質依存性に結合することを見いだした。この結合は、ホスファチジルセリン、ホスファチジン酸に特異的で、ホスファチジルコリン、ホスファチジルエタノルアミンでは認められなかった。カルスペクチンは、F-アクチンと結合し、F-アクチン架橋作用を有することが知られている。このカルスペクチンの機能へのアネキシンVIの効果を落下球法による粘度測定及び遠心法により調べた。アネキシンVIのCa及びリン脂質依存性のカルスペクチンへの結合が、カルスペクチンのF-アクチンへの結合を阻害し、カルスペクチンのF-アクチン架橋作用を阻害することが明かとなった。このように、アネキシンVIは、細胞膜直下で細胞膜骨格の消失、再構築といった細胞膜骨格機能制御におけるCaセンサーとして働き得る可能性が示された。
