新しいヘパリン親和性成長因子、プレイオトロフィンの神経栄養効果の解析

研究概要	プレイオトロフィン(PTN)の神経栄養効果の解析の一環として、PTNcDNA、抗PTN抗体を用いてPTNの遺伝子蛋白発現が大脳皮質損傷時の発現動態を検討した。大脳皮質障害後のPTNmRNAの発現は障害後1日より損傷部周囲に認められ、3日後にはピークに達した。この発現はその後暫減し、7日後まで認められたがその後は認められなかった。発現細胞は主に神経細胞であることが連続切片で観察したGFAP免疫染色のパターンとの比較により明らかとなった。一方MKmRNAの経時変化はほぼPTNmRNAのそれと一致していたが、発現の程度はPTNに比して弱かった。PTN、MKの蛋白発現の経時変化もそれぞれのmRNAのパターンとほぼ一致していたが、局在パターンには差異が認められた。すなわち損傷部に近接した皮質では両者ともに神経細胞体に強い免疫陽性反応が認められたのに対し、少しはなれた部位ではPTNが幼若期と同様の細胞膜に付随したパターンの免疫反応を呈するのに対し、MKではこのような染色は認められなかった。以上の結果はPTN、MK共に障害後の大脳皮質で早期に発現し、修復に関与していることを示唆すると考えられる。またPTN、MKの損傷遠隔部での局在の違いは、正常発生過程の結果と考え併せて、PTNが主に神経回路再構成に関与している可能性を示している。またバキュロウイルスベクターを用いてリコンビナントPTNを産生に成功した。精製リコンビナントPTNを用いた結合実験ではcoldPTNでブロックしうるバインディングが認められ、総結合量に対する非特異的結合量は約10-20%であった。現在解離定数、最大結合量、及びアフィニティークロスリンキングによる受容体の分子量などを検討中である。このことからPTNは非特異的に細胞膜面で突起伸長を促す分子ではなく、受容体を介した作用を持つ分子である可能性が高いと考えられる。
