研究概要	パラインフルエンザ2型ウイルスにおいて機能的に重要なP蛋白が産生されるためにはRNA-editingが必須のプロセスである。本研究では、ウイルスゲノムの転写及び複製をふまえたRNA-editing機構解析-Editingシグナルの同定-と本現象の普遍性を検討した。パラインフルエンザ2型ウイルス(PIV-2)のRNAレプリコン系の確立のためにPIV-2のP遺伝子とムンプスウイルスNP遺伝子の一部を結合した人工遺伝子を作製し、そのRNA転写物[-鎖]を細胞にトランスフェクトし、その蛋白の合成を抗ムンプスウイルスNPに対するモノクローナル抗体を用いて検出するという系を確立し、この系を用いてRNA-editing機構の解析を行なった。次にbaculovirus-vector系でPIV-2のP遺伝子を発現するとP蛋白のみならずV蛋白の合成がみられることを見い出し、baculovirus-vector系におけるRNA-editing様現象の詳細な解析を行なった。動物細胞においても、baculovirus-vector-Sf9細胞系で見られたRNA-editingが生じるかどうかをsRa-vectorを用いて検討したが、一時的な発現系でも構成的な発現系でも、RNA-editingは証明できなかった。このことはRNA-editingはbaculovirus-vector-Sf9細胞系に特有な現象であることを示唆しており、その系独自の因子を今後追究する予定である。PIV-2のV遺伝子およびSV41のV遺伝子をsRa-vectorをもちいてHeLa細胞で発現させたところ、V蛋白は核内に存在した。そのV蛋白上の核移行シグナルはC端側に存在することが明らかになったので、今後はその最小ドメインを決定する予定であるがV蛋白上には既存の核移行シグナルは見い出されないので、新しい核移行シグナルの発見につながるかもしれない。
