tRNAのアミノ酸受容特異性の分子識別

研究概要

tRNAのアモノアシル-tRNA合成酸素によるアミノ酸受容特異性の研究対象として、プロリンtRNAを選び、in vitro の系で検討した。その結果、大腸菌のプロリルヘtRNA合成酸素による認識部位として、アンチコドン2文字目のG35G36、アンチコドンに隣接するG37、識別位塩基A37、TPSIステムの中のG49、クセプタースムのC1-G72(特にG72)およびG3-C70塩基対、それにジヒドロアームの中のU17Aを同定した。プロノン化活性に影響を与えたこれらの部位真にプロリル-tRNA合成酸素によるポジティブな認識部位であることを確認するために、大腸菌のトレオニンtRNAにこれからの部位を順次導入する事により、ほぼ完全にプロリンを受容するように特異性のスイッチングが起こり、先に同定した部位の塩基対が真にプロリル-tRNA合成酸素により認識部位である事を立証した。また、大腸菌では唯一RNAレベルで一次構造が知られていない3種のプロリン tRNAの内、メジャーなプロリンtRNA(アンチコドン、CGG)のDNAプローブを用いて単離し、修飾を含めて一次構造の決定をおこなった。
