研究概要

細胞周期の観点から、細胞増殖・分化の制御機構を研究した。本年度は、(1)線虫C.elegansのフッ素イオン耐性変異遺伝子の1つflr-3の全長cDNAを分離し、これがATP結合部位の共通配列を持たないタンパク質キナーゼ(様分子)をコードしていることを明らかにした(桂)。(2)プロゲステロンやMosによる卵成熟の開始、およびMosによる未受精卵の分裂中期停止にMAPKK/MAPKカスケードが重要な役割を担っていることを明らかにした(西田)。(3)グリア酸性タンパク質やビメンチンの各リン酸化部位に特異的なモノクローナル抗体を用いて、分裂期の中間径フィラメントのリン酸化に、cdc2キナーゼの他にカゼインキナーゼが作動していることを明らかにした(稲垣)。(4)休止期の細胞はΔFosBの単独発現で細胞増殖を開始するが、この際G1/S期移行を規定するG1サイクリンとCDK2の発現が転写レベルの上昇ではなく各mRNAの安定化により起こることを明らかにした(中別府)。(5)出芽酵母タンパク質ホスファターゼPPIの変異のエンハンサー変異egp1を分離した。この遺伝子がコードする野性型タンパク質は、分裂酵母Sds22pと似ており、PP1と結合する(杉本)。(6)ヒト上皮細胞の増殖停止に特異的な遺伝子epi-1をクローニングし、この遺伝子が2種の転写産物を作ることを明らかにした(増井)。このように、多様な材料と手法を用いた研究を総合することにより、細胞増殖・分化の基本的なスイッチ機構と生物種による特殊性が明らかになると考えられる。
