研究概要

真核生物においてDNA損傷を中心とする遺伝情報の変化がもたらす細胞内のシグナル伝達機構や、DNA修復酵素系の働きを制御する仕組みを、細胞レベルで解明することを目的としている
1 HeLa細胞から8-ヒドロキシグアニン(oh^8Gua)の修復酵素の部分精製をした。哺乳類のN-メチルプリンDNAグリコシラーゼははoh^8Guaを除去修復することを見いだした(葛西)。
2 p53遺伝子をノックアウトしたトランスジェニックマウスより線維芽細胞を分離培養し、p53遺伝子欠損のDNA修復能への影響を調べた。p53欠損細胞では生存率に変化がなかったがSCEの頻度が増加した(石崎)。
3 DNA修復と関係するPCNAとXPACタンパク質との相互作用を解析し、D-サイクリンが結合することが知られているN-端(2-64)及びC-端(197-228)の2箇所に独立した結合することが見いだされた(松影)。
4 ゲルシフト法等を用いて、ヒトDNA損傷結合因子を解析して、XPE細胞に欠損する因子を見いだした。既に報告されているXPE-BFより分子量が小さいことから、異なる因子か検討中である(松永)。
5 XP患者の皮膚腫瘍よりDNAを抽出し、PCR-SSCP法により約50%にp53遺伝子の突然変異を見いだした。14の変異の内13は塩基置換変異であり、in vitroで見られる紫外線突然変異と傾向が同じであった(八木)
6 活性酵素によるDNA損傷を修復する大腸菌のmutMを相補するヒトやマウスの遺伝子を検索した。メチルプリングリコシラーゼがmutMのmutator活性を下げることを見いだした(山本)。
7 「多数細胞で条件付けられた培養液」からDNA修復を促進する活性物質は、分子量が1000以下で、酸性に弱く、活性炭にも吸着しなかったが、現在約100倍ほどに濃縮された。更なる精製を試みている(内海)。
