研究概要	本重点領域の計画研究は、情報転換器として機能する3量体GTP結合蛋白質(G蛋白質)が介在する情報伝達系について広範且つ集中的に研究することを目的として、計画班員12名により行われた。平成6年度おいては、数多くの細胞膜受容体と各種酵素・イオンチャネルとの共役のメカニズムについて、蛋白質の精製とcDNAクローニング、精製蛋白質を用いた再構成実験系、蛋白質をコードする遺伝子導入実験、蛋白質分子の詳細な構造と機能との関連などから検討された。1)新たにG蛋白質関連型の受容体、特に感覚受容器における受容体、松果体に存在する生物時計に関与する光受容体などのcDNAが多数単離された。また、脱感作現象と関連する受容体キナーゼによる受容体分子のリン酸化は、G蛋白質のβγサブユニットによって促進され、受容体分子の細胞内陥入と関連することが明かにされた。2)G蛋白質を介する効果器系の活性調節機構については、構成蛋白質の変異体や部分ペプチドなどを用いた検討から、蛋白質分子上の機能的な部位の解析が進められた。また、PI-3-キナーゼは、その特異的な阻害剤を用いた検討より、チロシンリン酸化とG蛋白質の両者によって制御される可能性が、さらに、ホスホリパーゼDの活性化に3量体のG蛋白質に加えて低分子量GTP結合蛋白質が関与するなどの新しい知見が得られ、G蛋白質の生理的役割の拡大に関する研究が進展した。3)G蛋白質分子の構造と機能との関連については、分子多様性をもつG蛋白質の構成サブユニットが精製され、αサブユニットへの脂肪酸付加やγサブユニットのプレニル化、リン酸化などの翻訳後修飾がサブユニット間の、あるいは受容体、効果器分子との相互作用に重要であることが明かにされた。4)NADから合成される新規環状ADPリボースのRIAが開発され、新たなセカンドメッセンジャーとしての機能解析が進められた。
