研究概要

シアル酸の分子種であるN-グリコリルノイラミン酸(NeuGc)発現制御に関与する因子の解析を行い、律速であるCMP-NeuAc水酸化反応には三つの蛋白性因子(cytochrome b5,NADH b5 reductase,末端水酸化酵素)が関与することを明らかにした。末端酵素を精製し、アミノ酸配列を明らかにし、cDNA解析も進行中である。今後、ヒトでの発現抑制の機序解明に興味がもたれる。腎細胞癌では硫酸化糖脂質が増加し、対応して硫酸転移酵素の活性が上昇する。また本酵素活性はEGFやTGFで活性が上昇し、この効果はCキナーゼを介するものであることが明らかになった。今後、Cキナーゼの活性化と酵素活性の上昇との関係に興味が持たれる。ガラクトース転移酵素は癌患者の血清中で、上昇している。本酵素に対する抗体を作成して、消化管の正常、癌組織を免疫組織染色すると、正常ではゴルジ体が染色されるのに、癌では細胞膜が染色された。癌組織の本酵素は正常よりも分子量の小さい分子が存在していることが明らかになった。子宮内膜上皮培養細胞で、コレステロール硫酸生合成に関与する硫酸転移酵素の活性化はプロゲステロンとその受容体の発現上昇によって制御されていることが明らかになった。コレステロール硫酸は子宮内膜に次いで、皮膚に高濃度に存在するが、その出現は多重層を形成する時期に一致し、ケラチン合成に先行することが明らかになった。癌関連抗原であるTn抗原はGalNAc-Ser/Thrが3残基連続して存在する構造であることが明らかになった。ほ乳類の腎臓に、これまでに報告されている動物レクチン(C,Sタイプ)に属さない、アネキシンファミリーに属する新しいタイプのレクチンを見いだした。リコンビナント蛋白の合成に成功し、今後リガンドの検索に興味が持たれる。
