研究概要	遺伝子トラップ法において、ES細胞を浮游培養して胚様体を形成させる方法がスクリーニング系としてすぐれていること、この方法により4つの遺伝子を単離したが、3つは既知の1つは未知の遺伝子であり、効率よくマウス内在性遺伝子を単離できることを明らかにした。YACおよびBACをマウス受精卵に注入する方法を確立した。δクリスタリン遺伝子の転写因子として単離されたδEF-1について、C末端側の破壊マウスでは主に胸腺の異常が出現するが、完全欠損ではnotochordの異常に基づく椎間板の欠損が起こることがわかった。始原生殖細胞の増殖にはgp130が関与すること、そのリガンドは未知のものであることが推察された。始原生殖細胞の増殖に関与する蛋白キナーゼであるskyのリガンドがGas6であること、それは精巣の間質細胞で発現すること、steel factoe存在下で増職能を有することを明らかにした。メダカの胞胚を培養することにより確立したOLES1およびOLES2細胞において、初期胚のマーカーであるSSEA-1やEMA-1が発現していることがわかった。ゼブラフィッシュの胞胚期の細胞はキメラ形成能のあること、卵黄がbFGF,Wnt8を介してOtxを抑制し、それが中胚葉誘導に関連することを明らかにした。ニワトリにおいてen-2が視蓋原基尾側への投射に関与するが、エレクトロポレーションによる視蓋領域への遺伝子導入法を開発し、それを用いてen-2の上流にPax5、下流にRagsおよびElf1があることを明らかにした。YACはリニアーであるが、環状化したベクターの開発に成功した。また、rad51株を用いることにより、キメラ率が5-10%に低下することを見いだした。
