標的遺伝子組換え法を用いたサイトカインの生理機能の解析

研究概要	1.β型及びγ型インターフェロン(IFN)遺伝子破壊マウス作製の試み標的遺伝子組換え用ベクターとして、ネオマイシン耐性遺伝子とチミジンキナーゼ遺伝子を用いたポジテイブ-ネガテイブ選択用のベクターと共に、IFNの発現を生体内で追跡できるようにlacZ遺伝子を組み込んだベクターも作製した。前者のベクターをE14もしくはA3-1のES細胞に導入し、β型あるいはγ型IFN遺伝子に相同組換えを起こしたESクローンを各々複数個単離した。これらのESクローンをマウスの胚盤胞に注入してキメラマウスを多数作製し、90%以上のキメラ率のものも得られたが、現在までのところ生殖系列への伝達が認められていない。親株のA3-1細胞については高い効率(5匹のキメラのうち3匹)で生殖系列への伝達が確認されたので、今後もA3-1細胞を中心にIFN遺伝子破壊マウスの作製を試みていく。 2.インターロイキン-1(IL-1)遺伝子破壊マウス作製の試み重要な炎症性のサイトカインであるIL-1遺伝子を破壊することも新しく始めている。相同遺伝子組換えを効率よく起こさせるためにES細胞と同じ129系統のマウス染色体遺伝子ライブラリーよりα型(14kb)及びβ型IL-1遺伝子(17kb)を単離した。α型IL-1遺伝子にはハイグロマイシン耐性遺伝子とlacZ遺伝子を挿入し、β型遺伝子にはネオマイシン耐性遺伝子とlacZ遺伝子を挿入した標的遺伝子組換え用ベクターを作製した。現在ES細胞に導入して相同組換えを起こしたESクローンの単離を試みている。
