研究概要

オーキシンによる伸長成長機構を明らかにするために、下胚軸にオーキシンを与えても偏差成長を起こさない突然変異体を一種類単離した。同変異体は地上部に特異的なオーキシン非感受性突然変異体であったので、オーキシン信号伝達機構は器官別に重複して存在していることが分かった。そこで、下胚軸特異的オーキシン非感受性を指標にして突然変異体を選択したところ、あらたに数種類の変異体を得た。これら変異体の分子遺伝学的研究によって、オーキシンの信号伝達機構を明らかにできると考えられる。オーキシン作用機構研究のもう一つの方向として、オーキシン誘導性遺伝子(ARG)の単離とその機能解析、およびARGプロモーターの誘導機構の研究を行った。本年度は下胚軸伸長成長系で新たに5種類のオーキシン誘導性遺伝子を単離したほか、ARG1と2については、プロモーター領域のクローニングを行った。ARGであるparBのプロモーター解析を行った結果、50から100bpの長さの二つのオーキシン応答性要素がオーキシン誘導に必要であることが分かった。また、もう一つのARGであるarcAの機能を明らかにするために、arcA産物と相互作用するタンパク因子をクローニングした。
シベレリン作用に関しては、アミラーゼ遺伝子のジベレリン応答シス配列を解析したほか、伸長反応系でジベレリン誘導性遺伝子を単離した。アズキ上胚軸から可溶性ジベレリン結合タンパクを2,200倍精製しリガンド特異性を調べたところ、各種ジベレリンとの間に高い活性相関があったことから、本標品の生理的意義が高いことが分かった。エチレン生合成調節機構に関しては、個々のACC合成酵素アイソザイムの発現レベルを調べるために特異抗体を作成したほか、同酵素遺伝子プロモーターとレポーター遺伝子の融合遺伝子による形質転換体を作成し、同遺伝子発現調節機構を解析する実験系を作成した。
