活性酸素代謝と関連遺伝子の発現制御機構解析

研究概要

人間生存のための地球本位型社会を実現するためには、生物の遺伝子に刻印された活性酸素代謝酵素系の制御機構とその生物学的インパクトを理解することが必要である。胎生期に低酸素の環境下(30-50mmHg)に生息する哺乳類は酸素ストレスから免れているが、生産に伴い約三倍(80-100mmHg)もの酸素濃度にさらされ、強い酸素ストレスを受ける。出産前SOD活性およびそのSODmRANは、脳、肝、腎とも低いが、出生後の成長にともなって活性が増加する。肺のみはSODの活性とmRNAの発現は出生の前後および成長に伴っても顕著な変化は見られなかった。成長に伴う遺伝子の発現は一酸化窒素合成酵素(NOS)でもみられた。従って、生物が高酸素濃度の環境内に移行する際には、酸素暴露によりSODの発現が増加することが明らかとなった。一方、Heme oxygenaseは、酸素ストレスに応答して出生直後から4日目までに著明に発現し、出生時の溶血で生じたヘム蛋白を有効に分解することが示唆された。ところで胎児期の肝臓は造血組織であるが、出生後は肝細胞が主となり、造血巣は骨髄に移行する。この肝細胞交代現象に出生時の酸素ストレスが関与する可能性が考えられる。事実、ラット肝臓よりミトコンドリアを採取し、呼吸調節能と電子伝達系の構成蛋白質群の発現を検討した結果、胎児ではコハク酸を基質とした酸素呼吸はほとんどみられず、出生後に呼吸を開始すると酸化的燐酸化反応が可能となってくる。出生前の胎児を帝王切開により、外因的に大気中に暴露させることによっても酸素呼吸を誘導することができる。電子伝達系構成成分のうち、胎児ではシトクロームbとシトクロームc oxidaseが非常に少ないことが知られているが、この蛋白群の発現も酸素ストレスに依存する可能性が示唆された。これらのことより、肝細胞の交代現象には出生時の酸素暴露が重要な役割をはたすことおよび活性酸素は病態増悪因子としてよりも生理活性物質としても作用している可能性が示唆された。
