研究概要	1.本研究は生物無機化学に芽生え次第に大きくなりつつある人体の化学および無機薬学への関心を集約し金属イオンの輸送と機能発現を軸に、生体分子空間に融合的な新物質群を用意し、そのダイナミズムを精査し、将来より精密な生体制御を目指した有機無機複合系の開発の基礎の構築を目的としている。 2.各班員により研究を展開した。矢野と加藤は分子識別能を有する前例のないCo(II)配糖錯体の合成に成功した。この系は対イオンによる錯体分子自身の構造変換が示唆され、酵素系におけるinduced fitの基本的なモデルの一つとして位置付けられる。青山は新しい金属酵素モデルとしての結晶内空孔の利用の可能性を見いだした。大倉はマウスに腫瘍細胞を移植し、腫瘍組織中および血漿中に含まれるポルフィリンを分析し、腫瘍組織にはプロトポルフィリンおよび亜鉛プロトポルフィリンが蓄積し、腫瘍の増大に伴い血漿中のプロトポルフィリンが増加することを明らかにした。大塚はNF-κBを介する細胞内シグナル伝達と転写の制御を目的とし、ラジカル消去作用をもつ化合物HPH-Pep、酸素活性作用をもつ化合物HPH、MeO-HPH、Me_2N-HPHを合成した。小谷は金属錯体と生体関連物質との間に働く弱い相互作用の構造依存性を明確にした。黒田は有機無機複合体によるDNAの分子認識の新たな方法を開発した。後藤は白金(II)錯体における芳香環-芳香環相互作用を解明した。北島は非ヘム鉄を含む蛋白質の活性点、特に酸素中間体の低分子量モデルの開発を目指し、種々の配位不飽和鉄(II)錯体の合成を行い、酸素との反応性を検討した。桜井は硫黄を配位原子にもつバナジウム(IV)錯体を合成し、糖尿病ラットに投与し、血糖値を正常化させることを見いだした。中井は光学活性ビナフトールと(i-PrO)_4Tiからえられるチタン錯体が優れた不斉触媒となることを見いだした。長野はsuperoxideを不均化して消去するsuperoxide dismutase活性を有する鉄錯体を合成し活性と構造の相関を検討した。西田は鉄-ntaおよび類似キレート錯体の構造と発ガン作用との関連性を明らかにした。廣瀬はDNAに結合した抗癌性白金錯体の立体構造を認識するモノクロナール抗体の作成を行った。千喜良と藤井は芳香環をもつ種々の銅(II)錯体が結合したDNAファイバーの錯体配位平面の配向性とESRを用いて錯体構造との関連を検討した。山村は生体適合性錯体の設計を目指して亜鉛および水銀オリゴペプチド錯体の構造を解明した。
