研究概要

最終年度に当たり、3年に亙り進めてきた研究の総合的なまとめを行ない報告書を作成した。特にα-アミラーゼ、β-アミラーゼ、グルコアミラーゼ、サイクロデキストリン合成酵素、α-グルコシダーゼ、プルラナーゼ等の酵素群の基質認識機構の比較、構造相関、新規機能性糖質合成への応用、産業化への可能性等について最終的なまとめを行ない、東京、熱海においてそれぞれ公開シンポジウムおよび最終報告会を行なった。
(1)糖質分解・合成酵素の構造および糖認識機構解析
α-アミラーゼ、β-アミラーゼ、グルコアミラーゼ、プルラナーゼ、α-グルコシダーゼ、セルラーゼ、サイクロデキストリン合成酵素、イソマルトデキストラナーゼ、β-グルコシダーゼなどの遺伝子工学的・化学的手法による一次構造やX-線による三次構造の解析、反応機構の解析を行った。一次構造の明らかとなった酵素については、すでに一次構造が明らかにされている糖質関連酵素とのアミノ酸配列の相同性の検討、その活性部位近傍のアミノ酸配列の比較を行ないこれらの酵素の活性部位における基質認識の機構について検討した。
(2)糖質分解・合成酵素の構造および機能改変
上記の酵素群について、遺伝子工学的・タンパク質工学的手法および化学修飾法により酵素反応に直接関与するアミノ酸残基とその位置を明らかにし、それらのアミノ酸残基を変えたsite-directed mutant 酵素を作製し、基質認識機構の解析を行い、より高度な利用を目的とした反応効率の高い酵素の作製を試みた。
(3)糖質分解・合成酵素の特異性解析および高度利用
新しく検索された酵素の反応機構および構造解析、それらの酵素群のもつ特異性、糖転移・縮合・水解作用による新規の機能性糖質の合成、開発を試みた。
