研究概要

光合成・光化学系IIの分子構築の解明にむけて、以下の解析が行われた。
(1)結晶化を目標に、好熱性ラン藻の数種について光化学系II標品を純化する方法を確立した。
(2)種々の架橋剤を用いて、光化学系II複合体を構成するサブユニット間や、光化学系II反応中心を構成するDI-D2タンパク質間の近接関係を明らかにした。また、化学修飾剤を用いる解析で、マンガンの結合に関係していると思われるヒスチジンやカルボキシル基の存在を明らかにした。
(3)パルスEPR法等により、電子伝達に関与する常磁性分子間の距離を決めるなど、構造的な新たな知見を得た。また、FTIR法により、Mn-クラスターとタンパク質、カルシウム、水分子等との相互関係を明らかにした。
P-680の三重項状態のFTIRによる解析から推定されたクロロフィル分子とD1タンパク質との水素結合を証明するため、Chlanydomonasを用いて形成転換株が作出された。
(5)光化学系II反応中心の結晶化条件を、界面活性剤の構造を中心に解析し、また、光化学系IIに関連する33kDaのタンパク質やアロフィコシアミンの結晶化を行い、後者についてはその精密な分子構造を決定した。
(6)光化学系II反応中心D1サブユニットのC-末プロセシングに関与する酵素の純化に成功し、その遺伝子の同定と塩基配列の決定を行った。また、単離したプロテアーゼとD1タンパク質のC-末に相当するオリゴペプチドを用いて、酵素の基質認識機構を解析した。
(7)D1タンパク質をコードする遺伝子(psbA)の特定の部位にin vitroでランダムな変異を入れ、強光下でスクリーニングすることにより、数種の光耐性シアノバクテリア(Synechocystis PCC 6803株)を得た。
