研究概要	中枢神経系、心血管などにおける薬物受容体とそのサブタイプの機能的役割や細胞内情報伝達系へのかかわりについて、異なった手法を用いた解析を行い以下の成果を得た。 1.中枢神経における受容体:慢性投与でエタノール依存症を獲得したマウス及び禁断症状を発現中のマウスでは、脳内ジアゼパム結合蛋白(DBI)mRNAの発現とDBI含量の増加が認められるが、これらにはベンゾジアゼピン受容体とエタノールの相互作用が関係する(栗山)。培養アストロサイトでエンドセリン(ET)_B受容体mRNAが発現する時ET-1によってMAPキナーゼが活性化されるが、この反応は百日咳毒素によって抑制されることから、同キナーゼ活性化に細胞内情報伝達系の関与が示唆される(後藤)。ラットの海馬において、α_2アドレナリン受容体を介するセロトニン遊離に百日咳毒素感受性のG蛋白の関与することがin vivoで示された(斉藤)。ラット線状体スライスにおいて、コリン性介在細胞の大型無棘細胞はドパミンによりD_2及びD_3の両レセプターを介して興奮し、中型有棘細胞はD_2レセプターを介して抑制されることが明らかにされた(笹)。高血圧ラットにおいて、バゾプレッシン産生細胞の室傍核にNK-3タキキニン受容体mRNAの発現を認め、この受容体の存在を確認した(神谷)。 2.心血管系における受容体:ウサギ胸部大動脈のα_<1A>-受容体サブタイプを介した収縮におけるCa^<2+>感受性増加機構にプロテインキナーゼC(PKC)が関与すること、PKCがボツリヌス毒素感受性低分子量G蛋白を活性化することを示した(高柳)。イヌ脳動脈におけるアセチルコリン反応に、内皮のムスカリン受容体刺激によるプロスタグランディン遊離を介する収縮と、拡張性神経のニコチン受容体刺激によるNOを介する弛緩が証明された(戸田)。ラット腸間膜動脈のNK-3受容体刺激は内皮依存性の血管拡張をひきおこす(神谷)。培養ヒト臍帯内皮細胞からのエンドセリン遊離は圧力負荷により増加するが、この反応はホスホリパーゼC阻害薬とCキナーゼ阻害薬により抑制することを示した(中木)。リゾホスファチジルコリンはモルモット心室筋細胞膜の非選択性陽イオン電流を増加し、カルシウムの流入を増加することにより細胞を障害することが結論された(安孫子)。 3.ホルモン受容体:甲状腺刺激ホルモン(TSH)受容体にはG蛋白共役型受容体キナーゼが発現しており、このキナーゼがTSH受容体の脱感作に関与することが示唆された(谷山)。
