研究概要	本研究計画の目標は、電気生理学(パッチクランプ、脂質平面膜)や分子生物学のエクスパートに、レーザー顕微分光学、クライオ電顕、分子動力学の専門家を加えて、イオンチャネルの動的構造に関する情報をいかに検出し、電気生理学的に得られた動的情報といかに結び付けるか、という課題に応えられる新しい研究の方法と戦略を創出することである。本年度は以下に列記する項目について成果が得られ、イオンチャネルの動的構造機能連関の本格的研究へ向けての方向と足場が確立した。 A)天然イオンチャネルの構造と機能 1)Ca依存性Kチャネルの調節機構(高井) 2)shaker K-チャネルのslowゲーティング機構(東田) 3)リアノジン受容体チャネルの透過機構(葛西) 4)内向き整流K-チャネルのゲーティング・ブロック機構(久保、老木) 5)チャネルゲーティングに対する脂質の効果(桐野) B)モデルチャネルの構造と機能 6)非ペプチド化合物による電位依存性チャネルの設計と解析(曽我部) 7)人工ペプチドチャネルの設計と構造機能連関(石田、曽我部) C)顕微鏡支援技術の開発 8)チャネル1分子の金コロイド標識による分子動態の検出(楠見) 9)1蛍光分子検出のための超低迷蛍光顕微鏡の開発(木下) 10)かごめ化合物とクライオ電顕のチャネル構造解析への応用(豊島)
