研究概要	脱分化過程が同調的に進行することでその機構解析に最も好適と考えられるタバコ葉肉プロトプラストの培養初期において、オーキシンにより発現誘導される遺伝子parA,parB,parCに関しては、parAの遺伝子産物が核に局在することを大腸菌に作らせたタンパク質に対する抗体で細胞を染色し、蛍光抗体法で観察することにより明らかにし、転写制御に関与している可能性を支持していることを明らかにした。また、parBに関する同様な検討によって、主として細胞質に分布し、特に色素体の周辺に分布することを共焦点レーザー顕微鏡観察で明らかにした。光親和性試薬アジドオーキシンとparAの遺伝子産物との結合からオーキシン結合タンパク質でにあることも示された。なお、脱分化状態の維持の条件の検討には最も好適な材料と考えられるタバコBY-2細胞におけるオーキシン除去および再添加による細胞分裂の誘導条件の確立とその条件の下で単離されてきたarcAに関して、構造解析及び発現制御の解析を行ない、arcAがGタンパク質のGβ様の遺伝子であり、細胞分裂と密接に関わって発現していることを明らかにした。更に、arcAの遺伝子産物を大腸菌に大量生産させることもできたので、抗体の産生及びそれを用いての細胞内局在の解析に道を開いた。研究課題の後半に関わる脱分化細胞よりの再分化に関しては、至適条件を見出すことができ、その条件で特異的に発現する遺伝子の候補も挙がっているので遺伝子の同定を急いでいる。
