環境ストレスが作物の窒素代謝に及ぼす影響の非破壊・細胞計測による解明

研究概要	植物細胞における非破壊計測のための基礎研究として、ダイズのモヤシ状の苗における細胞水分状態計測を行い、成長が起こっている細胞に特異的な水ポテンシャル場を発見し、非破壊状態における生理計測が生命現象を明らかにするためには不可欠であることが明らかにされた。細胞計測を行うさいに、細胞溶液を単細胞から植物生長を妨げることなく抽出する方法を開発することができたため、非破壊状態における生理代謝が明らかにできるようになった。計測法をニンジンの不定胚分化過程の計測に応用し、また水耕栽培におけるトマト植物を用いて計測を行った。塩ストレス下におけるトマト果実の尻腐れ現象に関連した代謝変化を調べると、細胞骨格成分とストレプタビジン結合蛋白質に特異的な変化が起こってきていることがわかった。また、ニンジンの不定胚分化を誘導するさいアンモニアイオン濃度を減少させ、硝酸イオン濃度を増加させることで不定胚分化能が増加することがわかっており、不定胚分化能力を持たないカルス群の分化ではストレプタビジン結合蛋白質の合成が起こらず、硝酸イオン濃度を増加させ不定胚分化能を持ったカルス群を分化させるとストレプタビジン結合蛋白質が特異的に分化されることが発見された。不定胚分化を起こると細胞間に膨圧極性がみられ、高い膨圧を持つ細胞群が根に分化を始め、低い膨圧を持つ細胞群が葉に分化を始めることが明らかにされた。胚がハート型ステージにはいるとストレプタビジン結合蛋白質の量が減少し、細胞骨格成分蛋白質の量が増えてくることから、窒素源の変化によって誘導された不定胚誘導カルスでのストレプタビジン結合蛋白質が細胞骨格形成とその後の形態形成を誘導する要因として重要であることが示唆された。
