研究概要

1.薬毒物動態の観点から腹膜透過性について検討し、11種類のバルビタール酸系薬剤の脂溶性と腹膜透過速度との間に相関関係が成り立つこと、並びにモノカルボン酸化合物の腹膜透過には特殊輸送系が存在する可能性を示唆する結果を得ている。現在、抗悪性腫瘍剤について検討中であるが,腹膜内に貯留傾向を示す薬剤や、腹膜に特殊輸送系が存在する可能性を示唆される薬剤についての研究結果も得られており、今後更に薬物の性状と腹膜透過性についての研究を進めてゆく予定である。
2.REMEDiシステム等を用いて,本年度は以下の点で新知見を得た.
(1)REMEDiシステムの充実・改善:昨年度に引き続きフェニトロチオン等計4種の薬物をライブラリーに新規追加した.更にヒドロキシジン等計10種の薬物の検量線を作成し,同時的に定性・定量できるようにした.更に、REMEDiシステムのパラメーターも検討し、これにより薬物同定の信頼性の向上及び迅速定量が可能になった。
(2)ボランティアによる服用実験:ジフェンヒドラミンをボランティアに服用させ血液、尿を分析した.測定値がジフェンヒドラミンの検出限界値(40ng/ml)より高く、薬物治療モニタリングの可能性が明らかになり、更に測定値をGC-MSのそれと比較した所、高い相関が得られ、REMEDiシステム有用性が示された。
(3)覚醒剤に関する実験的検討:法医学的に重要な覚醒剤のメタンフェタミン及びアンフェタミンと、腐敗した試料にしばしば発現するフェネチルアミンとの識別が重要である。REMEDiシステムではフェネチルアミンとアンフェタミンは識別不可能であったが、メタンフェタミンとは十分識別可能であり、更に、検出限界値は150ng/mlで覚醒剤メタンフェタミンの分析が可能であった。
