天疱瘡の発症機序の解明と新しい診断・治療法の開発

研究概要

尋常性天疱瘡抗原、落葉状天疱瘡抗原、デスモヨ-キンの複数の異なったドメインに対する抗体を用い、それらの正常人皮膚における微細局在部位を新鮮凍結組織超薄切片をもちいる免疫電顕法で同定した.すなわち新鮮凍結組織超微切片作成装置を用い、無固定、無包埋凍結固定皮膚から直径100nmの超薄切片を作成し、それを基室として金コロイドをマーカーとする免疫染色を施行した。それぞれのデスモゾーム抗原タンパクドメインの微細局在、および位置的相関関係については、それをラベルする金コロイドの分布、数をイメージアナライザー(computerized image analyzer)により測定し、統計処理により5nmレベルでタンパク個々の微細局在部位を決定した。
そして天疱瘡抗原タンパクのデスモゾームにおける微細局在部位と、実際の天疱瘡患者皮膚へのin vivoでの天疱瘡自己抗体の微細沈着部位を明らかにした。これにより天疱瘡の病変を惹起する抗原決定基の微細局在部位並びに、他のデスモゾーム構成タンパクとの位置的相関関係を解析した。
