研究概要	1.カエル神経筋シナプスについて、神経の断端からの軸索輸送によってシナプス前末端にCa^<2+>蛍光指示薬Indo-1を負荷することに成功した。UVレーザー共焦点顕微鏡を用いてmsオーターのCa^<2+>濃度変化の測定が初めて可能となった。100Hz、100回程度の高頻度で刺激すると神経末端内のCa2+濃度は細胞外が正常リンガー液の場合2μM程度に上昇し、刺激後は時定数400ms程度で急速に元に戻った。10〜20Hzの低頻度で刺激すると刺激中でもピークに達して減少することがあることがわかった。この結果、強力なCa^<2+>排出機構が存在すること、及び短期可塑性のうちの増進や増強は、単純な残存Ca^<2+>説では説明できないことが明らかになった。これは、今迄にザリガニや中枢シナプスで得られた結果と全くことなり、我々がCa^<2+>キレート剤BAPTAを負荷が短期可塑性に与える影響から推定したものに合致している。 2.カエル神経筋シナプスで見られる短期可塑性の中で寿命の短い促通の2つの成分速い促通と遅い促通の間には放出量に対して積の関係が存在することが明らかになった。また速い促通の減衰時定数は刺激に同期して起こる伝達物質放出であるEPPの場合と自発正の放出であるMEPPの場合で大きく異なり、前者は後者の約5倍であることが明らかになった。この結果は放出がシナプス前末端内のCa^<2+>濃度の6乗に比例して起こるとすると残存Ca^<2+>説でのみ説明することができる。即ち速い促通には古典的残存Ca^<2+>説が成立するがほかの3種の短期可塑性は異なる機構で起こることを示している。 3.イセエビ神経筋接合部のシナプスボタンの高速Ca^<2+>イメージングに成功し、ボタン内で神経刺激によって急速にCa^<2+>濃度が増大し、刺激ご急速に減衰するホットスポットが存在することが明らかになった。 4.イセエビの促通の2成分‘速い促通'と‘遅い促通'は共にBAPTA負荷によって減少した。これはカエルとは異なる促通の機構の存在を示している。
