研究概要	大脳皮質視覚野の神経回路の基本的構造は遺伝子により制御され、細部は学習的に形成されと考えられているが、その神経機構についてはほとんど明らかにされていない。平成8年度においては、ラット視覚野切片の長期増強の研究を行い、ラット視覚野にfLTPとsLTP2の種類のLTPが発現することを見出した(Kurotani et al. , 1997) 。 fLTPは早い時間経過で起こり、5分以内で最大となる。sLTPは時間経過が長く、20分-1時間で最大となる。sLTPには強い年齢依存性があり、成年ラットではほとんど発現しない。一方、fLTPは成年ラットで若年ラットの約2倍の頻度で発現する。若年ラットでsLTPとfLTPがほぼ同じ頻度で発現することを利用して、この2種類のLTPの分子機構の薬理学的研究を行った。sLTPはNimodipineなどの膜電位依存性Caチャンネル,Cycloheximidine、 Actinomycin Dなどの蛋白、RNA合成阻害剤により阻止され、一方,fLTPはAPVで阻止され、上記の薬物では阻止されない。また、2種類のLTPについて光学的記録を行い、sLTPが外側膝状体系の1次あるいは2次シナプスに柱状に発現するのに対し、fLTPは軸索側枝系に水平方向に発現する。これらの実験結果は2種類のLTPが全く異なる化学経路で異なる神経経路に発現することを示している。特にsLTPの発現に時間がかかり、蛋白あるいはRNA合成に依存し、外側膝状体系に発現し、一方、fLTPの発現が早く、蛋白あるいはRNA合成に依存せず、軸索側枝系に発現することが興味深い。また、このことはsLTPの役割は脳発達の神経回路形成に関与し、fLTPは情報処理のための短期記憶の役割を果していることを示唆している。
