薬物ターゲティングシステムの機能性評価法の確立と製剤特性-機能相関の体系化

研究概要

近年、薬物の体内挙動の制御を目的として、各種のDrug Delivery Systemの開発が活発に進められているが、なかでも薬物の体内分布過程を制御し、特定の標的部位に送達させるとともに作用部位以外の臓器への移行を回避することにより、薬物適用の最適化を図ろうとするいわゆる標的指向化(ターゲティング)の試みは、薬物治療の理想像を実現するものとして大きな注目を集めている。これを実現するための方法として、各種製剤学的技術を用いて優れた体内挙動特性を有する運搬システムが開発されているが、これらの研究の大部分は、標的指向化の視点からは試行錯誤的あるいは経験主義的に行われており、運搬体システムの設計、開発の基礎となる学問体系は今だ確立されていない。そこで本研究では、(1)標的指向化の評価解析法の確立、(2)運搬体システムの体内挙動特性の系統的整理、(3)標的指向化の方法論確立、の3点にわたって研究を展開した。標的指向化の評価解析法として、クリアランスの概念を基礎とし、マウスを用いて各種運搬体システムの全身動態特性を速度論的に解析する系を確立しその有用性を確認した。また、臓器および細胞レベルでの標的指向性を評価する方法として各種臓器潅流実験系および培養細胞を用いた実験系を確立した。以上の実験系を用いて、各種高分子あるいは微粒子性運搬体システムの体内動態を系統的に検討した結果、分子サイズや電荷等の物理化学的性質との関連を明らかにすることができた。「標的指向化(分布挙動制御技術)」のサイエンスとしての確立を図った。さらに以上の知見に基づき、抗酸化酵素superoxide dismutaseや高脂血症治療薬vitamin K5等のターゲティングシステムを開発することができた。
