機能性キラル化合物の不斉触媒合成

研究概要	不斉触媒反応は21世紀を間近に控えた現代有機合成化学の中心的課題である。また、機能性キラル化合物の効率合成はこの達成によって初めて成し遂げられる。本研究では、反応性に富み且つ極めて基本的な有機金属反応剤である有機リチウムを素材とし、触媒的不斉反応の開発とそのための基本原理の確立および医薬品候補物質等の機能性キラル化合物の不斉触媒合成を大きな目的とした。本年度は、概念的に新規な不斉触媒合成の第一歩として、有機リチウムを炭素求核剤とする触媒的不斉炭素-炭素結合形成反応、特に不斉1、4-及び不斉1、2-付加反応に的を絞って基本原理の発見・確立、さらにその分子論的基礎の構築を図ることを計画した。その結果、 1.鎖状α,β-不飽和エステルと有機リチウムの1、4-付加反応を触媒する不斉配位子の設計・応用を検討し、キラルヒドロベンゾインジメチルエーテルが触媒として機能し、付加生成物を良好な不斉収率で与えることを見いだした。 2.イミンへの不斉1、2-付加反応をキラルアミノエーテルが触媒し、90%以上の不斉収率が実現できた。 3.リチウムエノラートを求核剤とする光学活性β-ラクタムの不斉触媒合成の可能性を拓くことが出来た。
