ミトコンドリアDNAからみた現生人類の起源

研究概要

ヒト上科におけるミトコンドリアDNA(mtDNA)の塩基置換速度と分岐年代をより正確に推定するため、3人のヒト(アフリカ人、ヨーロッパ人、日本人)と3種のアフリカ類人猿(チンパンジー、ピグミーチンパンジー、ゴリラ)とオランウータンの全塩基配列を解析した。時間に対してほぼ直線的に蓄積している塩基置換を用いると、オランウータンとアフリカ類人猿は1,300万年前に分岐したという化石での推定年代のもとでは、ヒトとチンパンジーが490万年前に分岐したという結果が得られた。この分岐年代に基づいて、同義置換速度を推定したところ、3.89×10^<-8>/座位/年という値が得られた。Dループ領域における置換速度に関しては、7.00×10^<-8>/座位/年の値を得た。同義置換とDループ領域の置換の両方を用いることにより、ヒトmtDNAの最後の共通祖先の年代は143,000±18,000年前と推定した。アフリカ人の塩基配列が最も多様であるということと、上のヒトのntDNAの起源の年代より、現生人類ホモサピエンスのアフリカ起源説が強く支持された。
