先端成長細胞における成長点の誘導とその形成機構の解析

研究概要	植物の細胞伸長方式の一つは、細胞の一端のみで成長する先端成長である。この先端部には、植物細胞の伸長機構が集約されていると考えられる。先端部の構造と機能を解析するには、すでにできあがった先端部の観察よりも、先端部の分化・生成にともなった細胞内構造変化の時系列に沿った解析が望ましい。本研究では、シダ原糸体細胞の一部に新たな先端部を分化させ、その形成過程を観察する。今年度は、原糸体細胞を遠心し、赤色光を照射して成長点を誘導する系の改良と、細胞の遠心にともなう核の構造変化の観察を行なった。赤色光下で6日間培養した原糸体細胞に白色光で細胞分裂を誘導し、その後3日間赤色光下で細胞を伸長させた後、遠心して核を原糸体基部方向に移動する。この処理が最も成長点分化の確立が高いことが分かった。さらい核の移動距離に依存して成長点の発生率とその位置が制御されていることが分かった。原糸体の一部のみに赤色光を微光束照射すると、その近傍から成長点の分化が見られるが、微光束、核、成長点の三者の位置関係は必ずしも一致していない。細胞を基部方向に長時間遠心すると、核は原糸体基部方向に移動するのみならず、核マトリックスより比重の重い核小体は、核のDNA塊から離脱する。この結果、核本体と核小体は数百ミクロンのDNAの長い糸で結ばれた格好になる。このDNA糸は核膜に囲まれており、核が核本体と核小体の間で長く伸ばされたものである。このDNA糸はその後徐々に短くなり、再び核に吸収されるようだ。
