肝類洞壁細胞の動態に関する研究

研究概要

1)類洞周囲星細胞に対するKupffer細胞の作用
ビタミンAを貯蔵し、細胞外マトリックスを産生する星細胞を分離し、培養し、別に培養したKupffer細胞の培養上清を星細胞の培養基に加えると、星細胞の突起は、膜状から樹枝状に変化した。プロスタグランディンE_2を加えても同様な変化が起ったので、Kupffer細胞が放出するプロスタグランディンE_2によって類洞周囲の星細胞の形態が制御されていることが明らかとなった。生体内においては星細胞は特有な樹枝状を呈しているので、Kupffer細胞との機能的関係が存在することが示唆される。
2)肝類洞腔内に存在するクラスII陽性細胞とその動態
類洞腔内にはKupffer細胞とは別にクラスII抗原を発現している細胞が存在することを明らかにした。この細胞は類洞腔中のみならずDisse腔にも分布し、Disse腔中を移動し、グリソン鞘内のリンパ管に回収されることが示唆される所見をえた。この細胞の起源と清長について今後研究をすゝめたい。
