炎症反応発現における抑制基調サーボ機構の研究

研究概要	前年度までにこの実験に必要なウサギIL-lra遺伝子組換え体、およびそのポリクロナル抗体を開発し、本年度はこれに加えて中和活性のあるモノクロナル抗体を開発した。これに加えて以前に我々が開発したウサギIL-lβ遺伝子組換え体およびその抗体により、ウサギの系でこの1組のサイトカインの制御系を調べる道具が完全に揃えることができ、これらを用いてウサギ膝にLPSで起こした関節炎について以下の点を明らかにした。 1)この関節炎においてIL-lβの産生は6時間でピークとなり、その量は11.3±5.1fmol/関節で、抑制因子であるIL-lraの産生は9時間でピークとなり、量は2,121.8±824.1fmol/関節で、その産生量はモル比でみて、IL-lraが187倍も産生されている。2)IL-lβとIL-lraのtypeIレセプター(機能レセプター)への結合定数はほぼ等しい(Kd=350nM)。3)in vitroの胸腺細胞増殖誘導活性を指標にし、IL-lβの活性を完全に阻止するのに必要なIL-lraの量はモル比で100倍を要する。4)in vivoでIL-lβ2fmolにより誘導される白血球浸潤を指標にし、これをを完全に阻止するのに必要なIL-lraの量はモル比で10,000倍を要する。5)上述のLPS膝関節炎における白血球浸潤の抑制はIL-lra5.75pmolから検出されはじめ、575pmolで最も強い抑制が見られ、その抑制は70%に達するが、これ以上IL-lraの量を増やしても抑制は強くならない。6)このLPS炎症の白血球浸潤はIL-lraとともにTHFaに対する中和抗体を用いることにより達成され、その抑制は90%以上に達する。以上の結果から、IL-lβの作用は細胞上の極めて僅かのレセプターと結合することにより生物作用を発揮できること。さらに、炎症の場ではその初期からIL-lβという活性化因子と同時に大量のIL-lraを産生し、IL-lβの作用を局部化するような機構が存在していることを明らかにした。
