自己免疫病の発症に関写する遺伝子の解析・同定

研究概要

臓器特異的自己免疫病を自然発症するTCRα鎖トランスジェニックマウスモデルを用いて疾患感受性遺伝子を検索した。このモデルの特徴はTCRトランスジーンの導入によるT細胞異常のもとに、宿主の遺伝的背景によって様々な自己免疫病が発症することである。例えば、BALB/cでは胃炎が、A/Jでは卵巣炎がそれぞれ約80%の頻度で発症する。B10では頻度は共に0%である。胃炎、卵巣炎の感受性を規定するMHC,non-MHC遺伝子を検索するため、MHCコンジェニックマウスを用いてMHC遺伝子の関与を、マイクロサテライト遺伝子マーカーの検出による遺伝子マッピングによってnon-MHC遺伝子の関与を検索した。その結果、BALB.B(H-2b),BALB.K(H-2k)はBALB/c(H-2d)と同頻度で胃炎を発症したが、B10.D2(H-2d)の発症頻度は0%であった。
B10.A(H-2a),A.By(H-2b)の卵巣炎の頻度は0%であった。即ちH-2d,H-2k,H-2bは胃炎感受性であり、B10-non-MHC遺伝子中に胃炎抵抗性遺伝子が存在する。H-2bは卵巣炎抵抗性を規定する。またB10-non-MHC遺伝子中に卵巣炎抵抗性遺伝子が存在する。
(B10.D2xBALB/c)xBALB/c戻し交配マウスの胃炎、(AxBALB/c)F2での卵巣炎の頻度は、それぞれ27.6%,17.2%であり、それぞれの発症に複数個の遺伝子、恐らく2〜3個の遺伝子が関与すると推定された。胃炎、卵巣炎を発症したマウスについてマイクロサテライト遺伝子マッピングを行なった結果、胃炎感受性を規定する遺伝子の一つは第8染色体上に、胃炎/卵巣炎感受性遺伝子は第12、14染色体上にマップされた。また卵巣炎については第17染色体上MHC遺伝子の有意の関与が確認された。
