細胞内寄生菌のエスケープと宿主における感染防御免疫・組織傷害発現の機構

研究概要	細胞内寄生菌のマクロファージによる殺菌からのエスケープを可能にする病原因子が、感染宿主における細胞性免疫の誘導に関与する機構を解明し、さらに抗原特異的T細胞による免疫防御発現と組織傷害発現を、サイトカイン産生能を中心としたT細胞の機能や、その主要識別抗原の違いによって分離的に解析することを本研究の目的とした。マウスを用いた実験的感染モデルから、平成6年度には以下のような研究成果が得られた。 1.感染防御を担うIFN_<-γ>産生性CD4^+T細胞の識別抗原は、リステリアでは分子量の異なる幅広い抗原であり、IL‐2産生性T細胞の識別(反応)抗原のプロフィールとほぼ同様であった。ストレス蛋白など分泌性蛋白はあまり防御抗原としては作用していなかった。一方、M.bovis BCGではIFN_<-γ>産生性T細胞は主として18‐20kDaの蛋白のみを識別し、IL‐2が幅広い抗原に対して産生されるのと対照的であった。この抗原は菌体膜画分に多く、報告されている分泌抗原とは異なっていた。回復期結核患者末梢リンパ球もこの18kDa抗原によく反応してIFN_<-γ>を産生し、患者リンパ球のIFN_<-γ>産生能と回復の程度のは相関がみられた。2.リステリアのモデルでは、生菌免疫によって誘導されるCD8^+T細胞の受け身移入によっても若干の感染防御能がみられたが、細胞移入ではむしろ肝細胞傷害活性がみられ、この細胞が産生するTNFが組織傷害的に作用することが考えられた。3.感染早期の宿主で発現される各種サイトカインは、菌の病原性、特にリステリアではLLOに依存していた。感染早期に非特異的に産生されるサイトカインのうち、特にNK細胞由来のIFN_<-γ>は、マクロファージの細菌に重要なNOを作るiNOSの発現誘導に不可欠で、しかも生菌特有であった。
