研究概要	最近、我々はHLA-B35分子とHLA-B51分子のERから細胞表面までの輸送速度は、前者が早いのに比べて、後者は著しく遅いことを明らかにした。この違いはER内でのHLAクラスI分子とペプチドの結合力の差によって生じる可能性があるためHLA-B^3501分子とHLA-B^5101分子とペプチドの結合を測定する方法を確立し、これを用いてペプチドとHLA-B^3501分子、B^5101分子との結合能を測定した。HLA-B^3501およびB^5101結合自己抗原ペプチドのアミノ酸解析より明らかになったモチーフに一致した8-merから11-merの合成ペプチドを多数作製し、その結合能を調べたところ、HLA-B^3501結合ペプチドに比べてHLA-B^5101結合ペプチドの結合力は著しく低いことが明らかになった。このことからHLA-B35とHLA-B51の細胞内輸送の速度の違いは、粗面小胞体内でのHLA-B35およびHLA-B51分子とペプチドの結合力の違いによって生じていると推定された。また、HLAクラスI分子によってT細胞に提示する内因性ペプチドを明らかにするために、HLA-B51アロ反応性CTLクローンが認識するアロペプチドの分離を試みたところ、1つのCTLクローンが認識するTAP依存性のアロペプチドをHLA-B51分子から分離することができた。以前に明らかにしたヒトマイナ-組織適合抗原ペプチドの分離と合わせて、T細胞が認識する内因性ペプチドの分離方法を確立することができた。
