研究概要	リンパ球ホ-ミングの最も重要な段階であるリンパ球のリンパ節高内皮細胞(HEV)との相互作用には複数の接着分子を会した接着カスケードが存在することがこれまでの研究から強く示唆される。本研究ではこの接着カスケードのイニシエーター分子と考えられるL-セレクチンとそのリガンドについて主に研究をすすめた。その結果、次のようなことが明らかになった。 (1)可溶性組み替えL-セレクチンを用いてラットのリンパ節可溶化物からL-セレクチン結合物質を探索したところ、CD34(90kDa),GlyCAM-1(50kDa)のような既知のL-セレクチン・リガンドの他に180kDaの分子サイズをもつ未知のものと思われる物質の存在が確認された。(2)この物質を大量精製し、そのアミノ末端のアミノ酸配列を決定したところヒト肺胞マクロファージ由来のマンノースレセプターと90%以上の相同性が認められた。(3)得られた180kDaの物質は抗マンノースレセプター・ポリクローナル抗体と反応したが、この抗体はラット・リンパ節のHEVには反応せず、新しいL-セレクチン・リガンドとして同定された物質はHEV以外の部位に存在することが強く示唆された。(4)現在、この物質とL-セレクチンの相互作用の生物学的意義について検討中であるが、残念ながらその存在部位から考えて、リンパ球-HEV相互作用におけるカスケード反応には関与しない分子であると考えている。このほかに、L-セレクチンを介するシグナル伝達について解析を行ない、L-セレクチンを介して細胞内に特異的なシグナルが伝達されることを確認している。この分子は細胞内にわずか17アミノ酸残基しか持たないことから、シグナル伝達に必要な会合分子の存在が強く示唆される所見である。
