研究概要

ヒト精液のDNase I活性の起源は未だ不明である。そこで、諸臓器のDNase I活性の分布がヒトに類似しているウサギの生殖器系におけるDNase I活性分布を調査した。DNase I活性は精巣上体および精嚢に高く、それらの臓器のDNase Iを等電点電気泳動法で解析したところ、その泳動パターンは精液のパターンに類似していた。したがって、精液のDNaes Iは精巣上体および精嚢に由来する可能性が高いものと考えられた。
ヒトDNase Iのgenomic DNAの構造は不明である。ヒトゲノムDMAライブラリーからスクリーニングしたコスミドクローンなどを利用して、その全構造(ヌクレオチド配列)を明らかにしたところ、DNase I遺伝子は全長3.2kbであり、9個のエクソンから構成されていた。4種類の表現型1,2,3,4型のヌクレオチドレベルにおける相違を明らかにしたところ、1型と2型との間の相違はエクソン8の1塩基置換(A→G)によって生じたアミノ酸置換(Gln→Arg)に起因していること、3型は1型のエクソン6の1塩基置換(C→G)によって生じたアミノ酸置換(Pro→Ala)、4型は2型のエクソン2の1塩基置換(C→G)によって生じたアミノ酸置換(Gln→Glu)にそれぞれ起因していることが判明した。
精液フコシダーゼ(FUCA1)は優秀な個人識別マーカーであることが知られている。通常、かなりな量の白血球を使用して個人の型判定するので、法医鑑識分野での使用は極めて限定されていた。我々は、等電点電気泳動法に使用するゲルを改良し、微量な(5μ1)血清からFUCA1型を判定する方法の開発に成功した。
