ヒト消化管腫瘍におけるガストリン受容体遺伝子の発現とその突然変異に関する研究

研究概要

今年度の研究においては、私達が世界に先がけてクローニングしたヒト・ガストリン受容体cDNAを用いて以下の成績が得られた。
1.マストミスならびにヒトの胃カルチノイド腫瘍にガストリン受容体遺伝子の強い発現が認められた。
2.ヒト・ガストリン受容体遺伝子を導入したNIH3T3細胞では、ガストリン刺激によって細胞増殖が著明に促進され、その際Cキナーゼの活性化と同時にP42MAPおよびP125FAK(Focal adhesion kinase)のチロジン・リン酸化が促進された。さらにガストリンは本細胞のc-myc,c-fos遺伝子の発現を明らかに増殖させた。
3.ヒト肺小細胞癌細胞株の約50%にガストリン受容体遺伝子の発現が認められた。
4.ヒト大腸癌10例中2例、大腸癌細胞株8例中1例にガストリン受容体遺伝子の発現が認められた。
5.胃腺癌組織ならびに胃癌細胞株ではガストリン受容体遺伝子の発現が認められた例はなかったが、ヒト胃小細胞癌(内分泌癌)細胞株ECC10において強いガストリン受容体遺伝子の発現が認められた。
6.これらガストリン受容体遺伝子の発現している細胞株にガストリンを投与したところ、その細胞増殖が促進された。
7.これらガストリン受容体の発現している細胞からRT-PCR法によりガストリン受容体cDNAをクローニングし、このDNA塩基配列をみたところ、現時点ではガストリン受容体遺伝子の変異例は一例も見出していない。
