研究概要

1)脳ムスカリニックアセチルコリン受容体に特異的に結合するリガンド[N-^<11>Cメチル]-ベンゾトロピンを用いて健常人被験者7名についてポジトロンCTによる受容体の画像化を行った。また、受容体結合能定量のために、血液中の代謝産物の測定をHPLCを用いて行い、血液入力関数を求めた。次に脳の時間放射能曲線とこの入力関数を用いてパトラックプロット法により受容体結合能の定量を行った。また痴呆患者数人についてもポジトロンCTによる画像化を行った。さらに、各個人の脳の形態の違いによる差を標準化するために、コンピュータ脳図譜システム(HBA)を用いて、受容体分布画像を標準化し、7人の画像の平均値と標準と標準偏差の像をピクセル単位で作るべく作業中である。
2)アセチルコリンエステラーゼ(AchE)の代謝活性測定を目的として、^<14>C-N-メチル-3-ピペリジル酢酸を標識合成し、そのラット脳内における分布をオートラジオグラフィー法により画像化した。次にこの放射能分布と、組織化学的に測定した酵素活性分布とを比較して、両者の間に良い相関があることを明らかにした。また、N-メチル-3-ピペリジル酢酸のポジトロン標識化について種々の放射薬学的検討を行い、C-11による標識化に成功した。さらに、ポジトロンCTを用いてこのトレーサによるサル脳での画像化を初めて行った。その結果、線条体および視床において、放射能が高いことを明らかにした。現在、臨床応用開始のための準備(急性毒性の評価、被爆線量の評価、定量モデルの開発など)を進めている。
