精神分裂病の発症および経過に関与する脳内神経伝達物質の研究

研究概要

われわれが見出したドーパミンD2受容体遺伝子(DRD2)の311番目のコドンにおけるアミノ酸の置換(Ser→Cys)について、分裂病症例数を292名、対象者数を579名に増加して関連研究を行った。その結果分裂病では26名が(8.9%)がCys311を有しており、対象の24名(4.1%)より有意に多かった。Manchesterスケールで精神症状を評価し、陰性症状の有無で患者群を分けると、陰性症状のない82例中に14例(17.1%)ののCys311変異保有者を見出した。
この変異の機能的意義を解明するために、まずDRD2のshort form(D_<2S>)の311番目のコドンのシトシンをグアニンに組み換え、COS7細胞(一過性)とCHO細胞(安定性)に受容体蛋白を発現させた。受容体結合実験は^3H-スピペロンを標識リガンドとし、10μM(-)-スルピリド存在下の結合をベースラインとした。スキャッチャードプロットから算出したKd値は、Cys311型121μM、Ser311型78μMであった。各種アゴニスト、アンタゴニストの ^3H-スピペロンの結合阻害実験の結果、Cys311,Ser311それぞれに対するKiは、ハロペリドール4.7±1.5、2.6±0.3、ブロムペリドール1.4±0.5、2.4±1.8、クロザピン41±11、36±8.3、スルピリド13±8.5、20±12、ドーパミン110,000、60,000であり、両受容体の間に有意差はみられなかった。次に、培地にドーパミンを添加した時のdown regulationをみるため^3H-スピペロン結合を調べたが顕著な差はみられなかった。またinternalizationを測定するために^3H-スルピリドを用いたが、ドーパミン添加による差はみられなかった。
