人の卵胞発育とその病態に関与する卵胞内生理活性物質の作用機序

研究概要

卵巣機能、卵胞発育は、視床下部一下垂体一卵巣系のホルモン共に、局所の活性物質により調節制御されている事が近年明らかにされつつある。卵胞液には種々の生理活性物質が存在するが、我々は、ブタ卵胞液、ウシ卵胞液よりFSHを特異的に抑制するインヒビンを単離し、ウシ卵胞液よりFSH分泌を促進するアクチビンとその結合蛋白であるフォリスタチンを精製した。更に、ヒトFSHreceptor,ヒトLH/CGreceptorとラットactivin receptor typellのクローニングに成功している。卵巣局所で産生される生理活性物質のendocrine,paracrine,autocrine作用を検索することは、ヒトの卵胞発育機序の解明に重要であり、我々は、ヒト卵胞液よりの生理活性物質の単離精製、生理活性物質とゴナドトロピンレセプターの機能的な関係の解明を試みた。
ヒト卵胞液中のインヒビン濃度はブタ卵胞液、ウシ卵胞液のインヒビン濃度に比べ低濃度であり、その精製が困難であったが、モノクローナル抗体を用いた免疫吸着クロマト法の導入により単離精製に成功した。精製したヒトインヒビンを用い、更に精度の高いヒトインヒビンの測定系を開発中である。又、我々はアクチビンがゴナドトロピンレセプターの誘導を促進する事を報告しているが、今回、アクチビンが FSH receptor をmRNAレベルでも単独で強力に誘導する事、FSHとの協同作用によりLH recepotor をmRNAレベルでも誘導を促進する事を明らかにした。又、ヒトFSH receptor,ヒトLH receptor のCHO細胞での発現に成功し、分子生物学的手法によるdesensitization の機序解明を可能とした。アクチビンの結合蛋白であるフォリスタチンに関しては、アクチビンがフォリスタチン合成を最も強く促進する事、cAMPとPMSは共にフォリスタチン合成を促進する事を明らかとした。
