感覚受容の場としての歯根膜:終末装置機能特性解明の形態学的アプローチ

研究概要

1.実験的咬合性外傷時における歯根膜神経の変化:ラット臼歯を咬合挙上し、歯根膜神経の動態をprotein gene prduct9.5(PGP9.5)の免疫組織化学的手法用いて検討した。
(1)Ruffini神経終末の変化:咬合挙上2日では歯根膜Ruffini神経終末は膨化し、その輪郭が不鮮明になった。電子顕微鏡下では、軸索終末から長い原形質突起を伸ばしていた。また、軸索終末周囲のシュワン鞘に存在するcaveolaeの数が著しく減少した。この傾向は咬合挙上後7日目までみられた。
(2)細い神経線維の変化:咬合挙上2-4日にかけて圧迫側において数珠状を示す神経線維が有意に増加し、その後、経日的に減少傾向を示した。
歯根膜神経は咬合性外傷のような外的要因に特徴的な応答を示すことが明らかとなった。
2.Ruffini神経終末の発生に関する免疫電顕的検討:ラット切歯歯根膜Ruffini神経終末の発生過程をprotein gene product9.5(PGP9.5)による免疫電顕法を用いて検討した。Ruffini神経終末は多数のvesicleを含む小さな膨らみとしてその発生を開始した。Ruffini神経終末は咬合力によりその成熟過程が劇的に進行するが、そのさい神経成長円錐にみられるfilopodiaをおもわせる長い軸索終末の突起を伸ばすことが特徴であった。
3.顎反射系における神経調節物質の検討:ネコ咬筋にHRPを注入し、三叉神経運動核における神経調整物質の検討を行ったところ、HRPで標識された運動ニューロンはセロトニン陽性神経とシナプスを形成しており、セロトニンが顎運動調節に関与していることが示された。
4.歯根膜におけるNGF/NGFRの検索:研究代表者の交替と時間的制約のため十分な検討ができなかった。
