ストレス環境における口腔機能変化の生理学的解析

研究概要

慢性的なストレス環境下にある人では、無意識下の噛みしめや歯軋りなどの咀嚼筋活動が生じることは経験的に知られている。また、この現象の背後には、視床下部や大脳辺縁系など高次能の興奮による交感神経系の関与が考えられるが、そのメカニズムはほとんど明らかにされていない。本研究は、動物を用いて様々なストレスや抹消交感神経系の興奮が咀嚼筋活動に及ぼす影響を神経生理学方法を用いて解析することを目的とした。実験にはラットを用い、慢性実験と急性実験を行った。慢性実験では(1)ラットを4度の冷水中に入れる、(2)チューブの中に入れて拘束する、(3)無理な姿勢(立位)をとらせる、(4)不安薬(FG7142)をラットにあたえる等の操作を行った。しかし、いずれの場合でも咀嚼筋に著しい活動が生じなかった。また、麻酔を施したラットの顎関節腔内にマスタードオイルを注射して痛みを与えたが、一過性の閉口筋活動を除いては咀嚼筋に著しい活動が生じなかった。すなわち、不安や痛みのようなタイプのストレスは、長期間におよび閉口筋活動を生じさせないと言える。今後は、どのようなタイプの慢性的ストレスが有効であるかを調べる必要がある。
一方、麻酔したラットの頚部交感神経を刺激して閉口筋の活動を調べると、開口によって反射性に誘発された閉口筋活動(下顎張反射)は抑制された。この結果は予想に反するものであると言える。さらに、この抑制は閉口筋中の血流減少のためではなく、筋紡錘の活動が抑制されるために生じることが明らかとなった。また、交感神経の降下は中枢性に生じたものではなく、動脈に沿って閉口筋に入った末梢神経の活動によることが明らかにされた。なお、この研究はJ.Physiol.に掲載された。
